Low-fidelity DNA polymerases

چکیده

Also known as.... Translesion synthesis polymerases. There are two of them in Escherichia coli, polymerases IV and V, and five in human cells, polymerases ζ, η, ι, κ and Rev1 (so a knowledge of the Greek alphabet is essential when discussing them). Except for polymerase ζ, they fall into a new DNA polymerase family, designated the Y-family. Depending on the lesion, polymerase and context, they either insert the correct or an incorrect nucleotide opposite a damaged base. In the latter instances, they are responsible for generating mutations.

Error-prone DNA polymerases: when making a mistake is the only way to get ahead.

Cells have high-fidelity polymerases whose task is to accurately replicate the genome, and low-fidelity polymerases with specialized functions. Although some of these low-fidelity polymerases are exceptional in their ability to replicate damaged DNA and restore the undamaged sequence, they are error prone on undamaged DNA. In fact, these error-prone polymerases are sometimes used in circumstanc...

Specialized DNA polymerases, cellular survival, and the genesis of mutations.

Cell death caused by arrested replication of damaged or structurally altered DNA can be avoided in prokaryotic and eukaryotic cells by multiple DNA polymerases that are specialized to bypass DNA damage. Some of these polymerases perform such translesion DNA synthesis of specific types of damage with high genetic fidelity. However, they exhibit greatly reduced fidelity when they operate on undam...

An Overview of Y-Family DNA Polymerases and a Case Study of Human DNA Polymerase η

Y-Family DNA polymerases specialize in translesion synthesis, bypassing damaged bases that would otherwise block the normal progression of replication forks. Y-Family polymerases have unique structural features that allow them to bind damaged DNA and use a modified template base to direct nucleotide incorporation. Each Y-Family polymerase is unique and has different preferences for lesions to b...

DNA polymerases and somatic hypermutation of immunoglobulin genes.

Somatic hypermutation of immunoglobulin variable genes, which increases antibody diversity, is initiated by the activation-induced cytosine deaminase (AID) protein. The current DNA-deamination model posits that AID deaminates cytosine to uracil in DNA, and that mutations are generated by DNA polymerases during replication or repair of the uracil residue. Mutations could arise as follows: by DNA...

Subtle but variable conformational rearrangements in the replication cycle of Sulfolobus solfataricus P2 DNA polymerase IV (Dpo4) may accommodate lesion bypass.

The possible conformational changes of DNA polymerase IV (Dpo4) before and after the nucleotidyl-transfer reaction are investigated at the atomic level by dynamics simulations to gain insight into the mechanism of low-fidelity polymerases and identify slow and possibly critical steps. The absence of significant conformational changes in Dpo4 before chemistry when the incoming nucleotide is remo...

Translesion synthesis DNA polymerases and control of genome stability.

Eukaryotic and prokaryotic genomes are replicated with amazingly high fidelity to assure faithful transmission of genetic information from one generation to the next. The accuracy of replication relies heavily on the ability of replicative DNA polymerases to efficiently select correct nucleotides for the polymerization reaction and excise mistakenly incorporated nucleotides using their intrinsi...
